How much can the membrane change the result?

This application note compares VerdiBlot, PVDF and nitrocellulose in a fluorescence-based Western blot using a His-tagged recombinant protein in E. coli lysate.

To make the comparison meaningful, every membrane was tested with samples prepared from the same quantified lysate mixture. The proteins were separated under identical SDS-PAGE conditions, transferred by the same wet-transfer protocol and imaged using identical exposure settings within each fluorescence channel.

The study followed a loading series from 40 to 0.625 µg of total protein and evaluated:

• a 70 kDa His-tagged target protein
• a 150 kDa loading control
• signal intensity
• membrane background
• detectability at decreasing protein loads

Under the tested conditions, VerdiBlot showed the strongest overall performance and the highest sensitivity at low protein loads. The results also differed between the 70 kDa target and the 150 kDa loading control, showing why membrane performance should be assessed for the specific protein and imaging setup.
